  • 研究概述

    细胞内各个细胞器包括内质网、线粒体和胞浆都存在质量控制机制,破坏质量控制的任何环节都会影响细胞稳态,是细胞功能丧失。我们的实验室致力于以果蝇模型为主研究细胞器内质量控制的分子机制;各个质量控制系统之间的协调互作以维持细胞健康。

    1.线粒体运输协同线粒体自噬维持了神经元轴突中线粒体稳态,对神经元的生长和生理功能维持至关重要。围绕线粒体的质量控制机制,我们的实验室致力于揭示线粒体选择性顺行和逆行运输的分子机制;线粒体自噬及调控网络,并在果蝇、小鼠、以及人源细胞等多种研究模式中研究线粒体质量控制对神经元生理功能维持的生理意义。

    2.细胞磷脂代谢依赖于线粒体和内质网之间的脂质双向交换,多种磷脂如磷脂酰乙醇胺的缺乏引起多重应激信号。我们实验室致力于探索线粒体和内质网之间脂质双向交换的分子机制和生理功能,进而对细胞器之间如何协同反应以对抗各种压力。

    3.诸如多聚谷氨酰胺病等主要神经退行性疾病都与错误折叠蛋白质的聚集有关。我们实验室利用胞浆和内质网错误折叠蛋白质引发神经元退变的果蝇模型,深入探讨细胞如何感知错误折叠蛋白质,错误折叠蛋白在聚集前和聚集后的清除的机制,以及生物体如何在蛋白高度聚集时引发细胞死亡并清除异常细胞。
